LONG-TERM NICOTINAMIDE MONONUCLEOTIDE SUPPLEMENT INCREASES BLOOD NICOTINAMIDE ADENINE DINUCLEOTIDE LEVELS AND ALTERS MUSCLE FUNCTION IN HEALTHY OLDER MEN

Preclinical studies have revealed that an increase in Nicotinamide Adenine Dinucleotide (NAD+) with administration of nicotinamide mononucleotide (NMN) – an NAD+ precursor, may reduce aging-related disorders; however, human data on this are limited. So can oral nicotinamide mononucleotide supplementation increase blood NAD+ levels and alter physiological dysfunction in healthy older participants?


Aging-related diseases are associated with reduced NAD+ levels 

Aging is a risk factor for diabetes, cardiovascular disease, cancer and neurological diseases (e.g. Alzheimer's disease). Preventing physiological decline in old age is an important method to prevent aging-related diseases.

What is Nicotinamide adenine dinucleotide? 

Aging and age- related diseases have been shown to be closely related to reduced nicotinamide adenine dinucleotide – NAD+ levels. In animal studies, administration of intermediate NAD+ metabolites such as nicotinamide (NAM), nicotinamide mononucleotide (NMN) or nicotinamide riboside (NR) has been shown to increase NAD+ levels , leading to improved health. health and prolong the life of experimental animals. Therefore, the potential of NAD+ metabolites to improve tissue rejuvenation in humans has led to many clinical trials on NR and NMN.

  • NR – a substance similar to vitamin B3, is the main vitamin component in milk (~1 mg/L). 
  • NMN is found in foods such as edamame, broccoli and meat (~1mg/100g of food). However, because their concentrations in foods are very low, it is difficult to obtain sufficient quantities of these components from foods. 

Therefore, pure and concentrated NR and NMN have been used in clinical trials.

Administration of intermediate NAD+ metabolites has been shown to increase NAD+ concentrations

NR Clinical Trial Results: In these trials, NR (100–2000 mg/day) was administered to healthy participants or those with obesity for up to 12 weeks. Most NR clinical trials have reported the safety of NR use and shown elevated levels of NAD+ or NAD+-related metabolites in the blood. 

The most recent report found that NR increased fat-free muscle mass in obese participants, although no effects were observed on insulin sensitivity, mitochondrial function, and lipid accumulation in the liver and endothelium. planing.

In a 2020 study, the safety of oral administration of NMN was reported in humans for the first time. In addition, a double-blind, randomized, placebo-controlled, 10-week trial to evaluate the effect of NMN supplementation on metabolic function in 25 postmenopausal women with prediabetes was conducted. It was reported that NMN supplementation increased muscle insulin sensitivity , insulin signaling, and remodeling in overweight or obese women with prediabetes. 

Furthermore, the 2021 Study found that the effects of NMN supplementation combined with exercise were reported in healthy amateur runners aged 27–50 years. NMN dose-dependently increases ventilatory threshold and improves aerobic capacity during exercise.

To elucidate the safety and effectiveness of NMN use in older adults, scientists conducted a randomized, double-blind, placebo-controlled, parallel-group study using 250 mg NMN for healthy men 65 years of age and older for 12 weeks. 

In a previous study of NMN, a single oral dose of 500 mg of NMN and chronic administration of 250 mg of NMN for 10 weeks did not cause any specific harmful side effects. In the most recent study, long-term use of 1200 mg of NMN daily for 6 weeks did not cause any significant side effects. Likewise, chronic administration of 250 mg of NMN for 12 weeks in this study also did not cause any significant side effects. Thus, this study demonstrated that oral NMN supplementation at 250 mg/day in healthy older men for 12 weeks was safe, well-tolerated, and significantly increased concentrations of related metabolites. related to NAD+ and NAD+ in whole blood.

Furthermore, NMN administration partially improved muscle performance, as assessed by gait speed and grip strength in healthy older men. The researchers speculate that grip strength is only significant on the left side, as a significant difference can be made on the right side. Since ~90% of Japanese people are right-handed, the effects of NMN supplementation may be more pronounced in the left hand, which is less affected by daily exercise and movement.


Supplementing nicotinamide mononucleotide partially improves muscle performance
